Overview

This protocol describes DNA-sequencing of single-stranded DNA templates in reactions catalyzed by Sequenase (version 2.0). All the materials required for sequencing with Sequenase version 2.0 are contained in kits, sold by USB/Amersham Life Science, which include a detailed protocol describing the reagents and steps used in the sequencing reaction. This kit is invaluable for first-time or occasional sequencers, but it is expensive. After gaining experience, it may turn out to be more economical to purchase Sequenase version 2.0 in bulk and then assemble the remaining reagents locally. By Joseph Sambrook and David W. Russell
