Overview

Long double-stranded RNA (dsRNA) can be obtained after annealing sense and antisense RNA strands, or spontaneously during in vitro transcription of an inverted repeat. Sense and antisense DNA templates from a gene of interest can be prepared by PCR or from an appropriate plasmid carrying a piece of the gene and appropriate promoter for in vitro transcription using standard procedures.The following protocol describes in vitro transcription with the SP6 polymerase. For other polymerases (T3, T7), simply use the appropriate buffer and desired RNA polymerase instead of the SP6 polymerase. By Paula Stein and Petr Svoboda.
